Prophylactic Egcg Supplementation Attenuated The Dss
Given the benefit of green tea consumption in reducing the risk of IBD and different impacts on the gut microbiota by oral and rectal delivery of EGCG, prophylactic effect of EGCG on DSS-induced colitis was then explored. Mice were subjected to 2.5% DSS in drinking water for the last 6 days to induce acute colitis after receiving daily oral gavage dosed with 50 mg/kg body weight EGCG for 21 days , also as a simulation of about four to eight cups of tea per day for an adult . Disease symptoms and body weight were monitored daily. Mice were then sacrificed and the colonic pathology was evaluated. The concentrations of several representative inflammatory mediators in both the plasma and the colon were also measured. Apparently, prophylactic EGCG alleviated the symptoms of colitis in DSS-treated mice as indicated by significantly reduced daily DAI and body weight loss , and increased length of colon . Histological analysis further revealed that prophylactic EGCG suppressed DSS-induced infiltration of inflammatory cells and damage to the colonic mucosa . Moreover, prophylactic EGCG tended to reduce the plasma levels of IL-1 , IL-6 , IL-8 , and TNF- in mice with colitis. Strikingly, DSS-elevated levels of MPO , IL-6 , and TNF- in the colon were largely attenuated to normal levels by prophylactic EGCG.
These results collectively indicated that prophylactic EGCG was capable of suppressing DSS-induced colitis symptoms, colonic injury, and inflammation.
Grtp And Spontaneous Enterocolitis
We further examined efficacy of GrTP against enterocolitis in IL-10 deficient mice exposed to normal gut microbiota. Animals became anemic and showed increased colonic weight and splenic length due to accumulation of inflammatory cells and enterocolitis . IL-10 deficient animals tolerated Low and Mid doses of GrTP with significant improvement in their enterocolitis symptoms for the duration of experiment. While, animals on High dose lost weight and became moribund and were terminated. GrTP significantly improved anemia, decreased colonic weight , and normalized splenic length . In addition, IL-10 deficient mice had significantly high SAA and IL-1 compared to those kept at transgenic conditions. GrTP had no significant effect on IL-1 with a partial effect on SAA in this model .

IL-10 deficient mice became anemic with low hematocrit and GrTP significantly improved hematocrit in treated animals. IL-10 deficient mice had enlarged splenic tissue due to infiltration of inflammatory cells and GrTP significantly reduced the inflammatory response. IL-10 deficient mice developed spontaneous enterocolitis and rectal prolapsed. GrTP significantly ameliorated the pathological scores.
Isolation And Culture Of Lamina Propria Lymphocytes
LPL were isolated using a previously described method . Briefly, each cleansed colon was cut into 5-mm pieces and then incubated in 50 mL of PBS containing 0.1mol/L EDTA, 5 mmol/L dietythritol in a shaking water bath at 37°C for 30 min. The cultures were filtered and washed with PBS over a 100-mm sterile nylon mesh . The tissue fragments were then transferred to another flask and digested with 25 mL of complete RPMI supplemented with 1% collagenase Type IV at 37°C with agitation for 2 h. The resulting suspensions were filtered through a 100-m sterile nylon cell strainer . The cells were collected by centrifugation, and washed once with medium. The cells were resuspended in 5 mL of 45% Percoll , layered onto 65% Percoll and centrifuged at 600 × g for 20min at 20°C. Lymphocytes were removed from the interface of 4565%, washed and counted. The resulting LPL were cultured in 1.0 mL of complete RPMI for 24 h under the conditions described above. Culture supernatants were collected and stored at 70°C.

The odds ratio for green tea consumption for diabetic retinopathy patients was 0.49 . When stratified by sex, the green tea consumption and protective effect of green tea on retinopathy was more significant in female than male participants . When adjusted for age and sex, green tea consumption was reported to be significantly associated with reversed diabetic retinopathy , high systolic blood pressure , duration of diabetes , and the presence of family history of diabetes . Therefore, those diabetic patients who regularly consumed green tea had a significant retinopathy risk reduction of about 50% compared with those who had not.

At the end of 1 wk, male mice from each group were humanely killed and colon explant and LPL cultures established. The remaining 14 mice in each group continued to receive treatment for 6 wk. As shown in , both the colon explants and LPL from placebo-treated IL-2/ mice spontaneously secreted IFN-. In contrast, LPL isolated from wild-type mice did not secrete detectable amounts of IFN- . Compared with sucrose-treated mice, GrTP-treated mice demonstrated a significant reduction in the spontaneous release of IFN- from colon explants and LPL . TNF- release in explant cultures from mice treated with GrTP was also significantly diminished in the explant cultures, but there was no noted difference in the LPL cultures. The lack of a difference in the LPL cultures probably reflected the low concentrations measured and suggests that lymphocytes may not be the major source of TNF- in the lamina propria. These data show that after 1 wk of ingestion of GrTP, there was a reduction in IFN- and TNF- production by the inflamed mucosa.

Total microbial genomic DNA of each fecal sample was extracted using QIAamp DNA Isolation Kit .
